Tau蛋白对维持神经细胞内的微管结构的稳定发挥了重要作用。在阿尔茨海默病(AD)中,Tau蛋白的异常过度磷酸化会显著促进神经退行性过程。研究发现,相比于β淀粉样蛋白斑块,磷酸化Tau蛋白更适合作为AD疾病进展和神经纤维病理学的指标。
高度磷酸化的Tau蛋白(p-tau)组成的神经纤维缠结是AD的标志性病理之一。但是Tau蛋白发挥稳定微管的作用,是神经元发挥功能必不可少的。只有当Tau蛋白发送异常磷酸化,不断聚集并形成纤维缠结时,才会成为生成AD的“凶手”。近期的研究还发现,Tau蛋白是阻止AD发生的关键成分,可有效帮助神经胶质细胞清除毒性的脂质,减少对神经元的损伤。
